t is now well accepted that defined architectural compartments within the cell nucleus can regulate the transcriptional activity of chromosomal domains within their vicinity. However, it is generally unclear how these compartments are formed. The nuclear periphery has received a great deal of attention as a repressive compartment that is implicated in many cellular functions during development and disease. The inner nuclear membrane, the nuclear lamina, and associated proteins compose the nuclear periphery and together they interact with proximal chromatin creating (...) a repressive environment. A new study by Harr et al. identifies specific protein–DNA interactions and epigenetic states necessary to re‐position chromatin to the nuclear periphery in a cell‐type specific manner. Here, we review concepts in gene positioning within the nucleus and current accepted models of dynamic gene repositioning within the nucleus during differentiation. We report the identification of a recurrent, 520-kb 16p12.1 microdeletion associated with childhood developmental delay. The microdeletion was detected in 20 of 11,873 cases compared with 2 of 8,540 controls and replicated in a second series of 22 of 9,254 cases compared with 6 of 6,299 controls. Most deletions were inherited, with carrier parents likely to manifest neuropsychiatric phenotypes compared to non-carrier parents. Probands were more likely to carry an additional large copy-number variant when compared to matched controls. The clinical (...) features of individuals with two mutations were distinct from and/or more severe than those of individuals carrying only the co-occurring mutation. Our data support a two-hit model in which the 16p12.1 microdeletion both predisposes to neuropsychiatric phenotypes as a single event and exacerbates neurodevelopmental phenotypes in association with other large deletions or duplications. Analysis of other microdeletions with variable expressivity indicates that this two-hit model might be more generally applicable to neuropsychiatric disease. © 2010 Nature America, Inc. It is widely recognized that ‘global’ symmetries, such as the boost invariance of classical mechanics and special relativity, can give rise to direct empirical counterparts such as the Galileo-ship phenomenon. However, conventional wisdom holds that ‘local’ symmetries, such as the diffeomorphism invariance of general relativity and the gauge invariance of classical electromagnetism, have no such direct empirical counterparts. We argue against this conventional wisdom. We develop a framework for analysing the relationship between Galileo-ship empirical phenomena on the one hand, and (...) physical theories that model such phenomena on the other, that renders the relationship between theoretical and empirical symmetries transparent, and from which it follows that both global and local symmetries can give rise to Galileo-ship phenomena. Entire moral philosophies have been rejected for ruling out the possibility of failure. This “fallibility constraint” (also sometimes called the “error constraint”) cannot be justified by appealing either to Wittgensteinian considerations about rules or to the moral importance of alternate possibilities. I propose instead that support for such a constraint in ethics can be found in the Strawsonian reactive attitudes. I then use the constraint to reveal hidden weaknesses in contemporary contstitutivist strategies to ground moral normativity such as Christine Korsgaard’s, (...) and also to reveal hidden strengths in historical accounts of morality such as Bishop Butler’s. We will have reason to reject any moral theory that makes constitutivism’s mistake, but only because we have reason not to reject the fallibility constraint itself. The way this ethical fallibility can be justified suggests a general principle that could be used to justify fallibility constraints in other normative domains such as practical reason, epistemology, the philosophy of language, and the philosophy of mind. Consisting of fewer than two hundred verses written in an obscure if not impenetrable language and style, Patanjali's Yoga Sutra is today extolled by the yoga establishment as a perennial classic and guide to yoga practice. As David Gordon White demonstrates in this groundbreaking study, both of these assumptions are incorrect. Virtually forgotten in India for hundreds of years and maligned when it was first discovered in the West, the Yoga Sutra has been elevated to its present iconic (...) status--and translated into more than forty languages--only in the course of the past forty years. White retraces the strange and circuitous journey of this confounding work from its ancient origins down through its heyday in the seventh through eleventh centuries, its gradual fall into obscurity, and its modern resurgence since the nineteenth century. Tracing the remarkable trajectory of this enigmatic work, White's exhaustively researched book also demonstrates why the yoga of India's past bears little resemblance to the yoga practiced today. Over the past fifty years, a number of approaches to the recovery of the multiple pasts of Hinduism have held the field. These include that of the discipline of History of Religions as it is constituted in North America as well as those of the Hindu nationalists, the col and post-colonial historians, and the Subaltern Studies School. None of these approaches have proven satisfactory because, for methodological or ideological reasons, none have adequately addressed human agency or historical change in their (...) accounts of the pasts out of which modern-day Hinduism has emerged. The Hindu nationalist historians hark back to an extended Vedic golden age in which religious practice remained unchanged until the corruptions spawned by the Turkish invasions of the eleventh century. Many Western indologists and historians of religion specializing in Hinduism never leave the unalterable ideal worlds of the scriptures they interpret to investigate the changing real-world contexts out of which those texts emerged. The colonial and postcolonial historians focus on the past two hundred years as the period in which all of the categories through which India continues to interpret itself—including Hinduism—were imposed upon it from without. Adducing examples of Hindu practitioners and thinkers from the colonial period, subaltern theorists and others argue that historical thought is itself alien to the authentic Indian mind. This article suggests a number of interpretive strategies for retrieving the multiple Hinduisms of the past and of the medieval period in particular as that time out of which most modern-day practices of Hinduism emerged. These include an increased emphasis on non-scriptural sources and a focus on regional traditions. (shrink)

Five medieval Sanskrit-language descriptions of a fabulous technique for extracting mercury from the “wells” in which it naturally resides are shown to be remarkably similar to accounts preserved in Chinese and Syriac. Whereas the Sanskrit and Chinese versions date from no earlier than the thirteenth century C.E., the Syriac version dates from no later than the tenth century. The present article first compares and contrasts these three alchemical narratives, and then suggests that all three are perhaps related to a broader (...) and far more ancient Indo-European mythic tradition of a deity associated with the phenomenon of “Fire in Water,” as attested in Vedic, Avestan, Roman, Irish, and Greek sources. All eight of these witnesses appear to attest to ancient religious and scientific traditions relative to geothermal phenomena. (shrink)

As David White explains in the Introduction to Tantra in Practice, Tantra is an Asian body of beliefs and practices that seeks to channel the divine energy that grounds the universe, in creative and liberating ways. The subsequent chapters reflect the wide geographical and temporal scope of Tantra by examining thirty-six texts from China, India, Japan, Nepal, and Tibet, ranging from the seventh century to the present day, and representing the full range of Tantric experience--Buddhist, Hindu, Jain, and even (...) Islamic. Each text has been chosen and translated, often for the first time, by an international expert in the field who also provides detailed background material. Students of Asian religions and general readers alike will find the book rich and informative. The book includes plays, transcribed interviews, poetry, parodies, inscriptions, instructional texts, scriptures, philosophical conjectures, dreams, and astronomical speculations, each text illustrating one of the diverse traditions and practices of Tantra. Thus, the nineteenth-century Indian Buddhist Garland of Gems, a series of songs, warns against the illusion of appearance by referring to bees, yogurt, and the fire of Malaya Mountain; while fourteenth-century Chinese Buddhist manuscripts detail how to prosper through the Seven Stars of the Northern Dipper by burning incense, making offerings to scriptures, and chanting incantations. In a transcribed conversation, a modern Hindu priest in Bengal candidly explains how he serves the black Goddess Kali and feeds temple skulls lentils, wine, or rice; a seventeenth-century Nepalese Hindu praise-poem hammered into the golden doors to the temple of the Goddess Taleju lists a king's faults and begs her forgiveness and grace. An introduction accompanies each text, identifying its period and genre, discussing the history and influence of the work, and identifying points of particular interest or difficulty. The first book to bring together texts from the entire range of Tantric phenomena, Tantra in Practice continues the Princeton Readings in Religions series. Our understanding and praxis of spiritual formation is only as sound as our theology of the Holy Spirit. Unfortunately, there is a common shortcoming that leaves our theology of the Spirit less than fully capable of rendering an adequate account of the Holy Spirit's agency in transforming a person into the likeness of Christ. Theology of the Holy Spirit commonly emphasizes victorious and genteel themes while failing to articulate adequately that the Spirit is a direct agent in transformative suffering. Some (...) of the Comforter's most important sanctifying work is exceedingly discomforting. This article focuses on these less palatable aspects of the Spirit's work in an attempt to tell a fuller story about the Spirit. By adding often-neglected dimensions and textures to our theology of the third Person, we are more capable of articulating the full range of the Spirit's role in formation. (shrink)

This paper summarizes a discussion session investigating the corporate representatives behind corporate citizenship and sustainability initiatives.

Public acceptance is often seen as a key reason why water-recycling technology is rejected. A common assumption is that projects fail because the general public is unable to comprehend specialist information about risk and the belief that if the public were better informed, they would accept change more readily. This article suggests that rhetoric about acceptance is counterproductive in progressing sustainability as it does not address issues relating to institutional arrangements and reinforces a dichotomy between expert and lay groups. Instead, (...) it is argued that institutional change is needed to build opportunities for constructive public engagement. The failure to implement sustainable water use through recycling can be understood as the result of several factors including present cost structures for water, institutional conservatism, administrative fragmentation, and inadequate involvement of communities in planning. Achieving sustainable water use through recycling may require better coordination between agencies and integrated government policies. (shrink)

Is there progress in philosophy? A glass-half-full view is that there is some progress in philosophy. A glass-half-empty view is that there is not as much as we would like. I articulate a version of the glass-half-empty view, argue for it, and then address the crucial question of what explains it.

Addressing fake news requires a multidisciplinary effort The rise of fake news highlights the erosion of long-standing institutional bulwarks against misinformation in the internet age. Concern over the problem is global. However, much remains unknown regarding the vulnerabilities of individuals, institutions, and society to manipulations by malicious actors. A new system of safeguards is needed. Below, we discuss extant social and computer science research regarding belief in fake news and the mechanisms by which it spreads. Fake news has a long (...) history, but we focus on unanswered scientific questions raised by the proliferation of its most recent, politically oriented incarnation. What ontology does realism about the quantum state suggest? The main extant view in contemporary philosophy of physics is wave-function realism . We elaborate the sense in which wave-function realism does provide an ontological picture, and defend it from certain objections that have been raised against it. However, there are good reasons to be dissatisfied with wave-function realism, as we go on to elaborate. This motivates the development of an opposing picture: what we call spacetime state realism , a view (...) which takes the states associated to spacetime regions as fundamental. This approach enjoys a number of beneficial features, although, unlike wave-function realism, it involves non-separability at the level of fundamental ontology. We investigate the pros and cons of this non-separability, arguing that it is a quite acceptable feature, even one which proves fruitful in the context of relativistic covariance. A companion paper discusses the prospects for combining a spacetime-based ontology with separability, along lines suggested by Deutsch and Hayden. (shrink)

Platonists affirm the existence of abstract mathematical objects, and Nominalists deny the existence of abstract mathematical objects. While there are standard arguments in favor of Nominalism, these arguments fail to account for the necessity of Nominalism. Furthermore, these arguments do nothing to explain why Nominalism is true. They only point to certain theoretical vices that might befall the Platonist. The goal of this paper is to formulate and defend a simple, valid argument for the necessity of Nominalism that seeks to (...) precisify the widespread intuition that mathematical objects are somehow ‘spooky’ or ‘mysterious’. We start by presenting three different views that jointly imply that every person has many conscious beings in their immediate vicinity, and that the number greatly varies from person to person. We then present and assess an argument to the conclusion that how confident someone should be in these views should sensitively depend on how massive they happen to be. According to the argument, sometimes irreducibly de se observations can be powerful evidence for or against believing in metaphysical theories.

Adam Ferguson has received little of the renewed attention that contemporary philosophers have given to the philosophers of the Scottish Enlightenment, most notably David Hume, Thomas Reid and Adam Smith. There are good reasons for this difference. Yet, the conception of moral philosophy at work in Ferguson's writings can nevertheless be called upon to throw important critical light on the current enthusiasm for philosophical ethics and applied philosophy. The witness of the Scriptures and of virtually every human culture suggests that one of the most pivotal and thus crucial dimensions of human formation, and thus spiritual formation, is the intergenerational dynamic: older men with younger men; older women passing on the faith to younger women. One generation encouraging, blessing and transmitting wisdom to the next generation. And yet, it is often observed that this is a missing dimension of congregational life–that congregations are increasing stratified along generational lines. Thus (...) this essay will make the case that the church is a liturgical, catechetical and missional community and that faith formation, in Christian community, will happen through the interplay of the generations in worship, teaching/learning and service. And that therefore congregations would be wise to be more attentive to how they foster intergenerational connections, rather than inadvertently or intentionally create generational divides in the church. Also, it is important to stress that a critical part of congregational life is that of equipping older men and women to be a means of grace to their younger counterparts. The essay will draw on the Biblical references to intergenerational formation, the insights of contemporary social-psychology and the observations of the author regarding contemporary congregational life. Given their physical realization, what causal work is left for functional properties to do? Humean solutions to the exclusion problem (e.g. overdetermination and difference-making) typically appeal to counterfactual and/or nomic relations between functional property-instances and behavioural effects, tacitly assuming that such relations suffice for causal work. Clarification of the notion of causal work, I argue, shows not only that such solutions don't work, but also reveals a novel solution to the exclusion problem based on the relations between dispositional properties at (...) different levels of mechanism, which involves three central claims: (i) the causal work of properties consists in grounding dispositions, (ii) functional properties are dispositions, and (iii) the dispositions of mechanisms are grounded in the dispositions of their components. Treating functional mental properties as dispositions of components in psychological mechanisms, I argue that such properties do the causal work of grounding agent-level dispositions. These dispositions, while ultimately grounded in the physical realizers of mental properties, are indirectly so grounded, through a hierarchy of grounding relations that extends upwards, of necessity, through the mental domain. According to dispositionalism about modality, a proposition <p> is possible just in case something has, or some things have, a power or disposition for its truth; and <p> is necessary just in case nothing has a power for its falsity. But are there enough powers to go around? In Yates (2015) I argued that in the case of mathematical truths such as <2+2=4>, nothing has the power to bring about their falsity or their truth, which means they come out both (...) necessary and not possible. Combining this with axiom (T), it is easy to derive a contradiction. I suggested that dispositionalists ought to retreat a little and say that <p> is possible just in case either p, or there is a power to bring it about that p, grounding the possibility of mathematical propositions in their truth rather than in powers. Vetter’s (2015) has the resources to provide a response to my argument, and in her (2018) she explicitly addresses it by arguing for a plenitude of powers, based on the idea that dispositions come in degrees, with necessary properties a limiting case of dispositionality. On this view there is a power for <2+2=4>, without there being a power to bring about its truth. In this paper I argue that Vetter’s case for plenitude does not work. However, I suggest, if we are prepared to accept metaphysical causation, a case can be made that there is indeed a power for <2+2=4>. (shrink)

We examine two different descriptions of the behavioral functions of the hippocampal system. One emphasizes spatially organized behaviors, especially those using cognitive maps. The other emphasizes memory, particularly working memory, a short-term memory that requires iexible stimulus-response associations and is highly susceptible to interference. The predictive value of the spatial and memory descriptions were evaluated by testing rats with damage to the hippocampal system in a series of experiments, independently manipulating the spatial and memory characteristics of a behavioral task. No (...) dissociations were found when the spatial characteristics of the stimuli to be remembered were changed; lesions produced a similar deficit in both spatial and nonspatial test procedures, indicating that the hippocampus was similarly involved regardless of the spatial nature of the task. In contrast, a marked dissociation was found when the memory requirements were altered. Rats with lesions were able to perform accurately in tasks that could be solved exclusively on the basis of reference memory. They performed at chance levels and showed no signs of recovery even with extensive postoperative training in tasks that required working memory. In one experiment all the characteristics of the reference memory and working memory procedures were identical except the type of memory required. Consequently, the behavioral dissociation cannot be explained by differences in attention, motivation, response inhibition, or the type of stimuli to be remembered. As a result of these experiments we propose that the hippocampus is selectively involved in behaviors that require working memory, irrespective of the type of material that is to be processed by that memory.
